Title: A UV-Visible-NIR, Time-Resolved Fluorescence Spectrometer for High-Pressure Biological Studies

Description: A high-pressure, time-resolved fluorescence spectrometer has been designed and characterized. The system consists of a nitrogen-pumped dye laser for UV-visible–NIR excitation (337.1-960nm wavelength), a pressure pump (maximum 200MPa), a quartz capillary pressure cell, an avalanche photodiode (APD), a 1-GHz digital oscilloscope, a 1-nm-resolution spectrometer and a fast-gated intensified charge coupled device (ICCD). The system is characterized as a spectroscopic probe at high pressures. A high-pressure cell has been designed and characterized to a stable pressure of 80MPa. The spectrofluorometer has been calibrated and tested for flatfieldness, linearity of signal with gain, and linearity of signal with excitation power. Spectra have been collected from samples at atmospheric pressure and at elevated pressures. Lifetime information has been obtained from samples at atmospheric pressure and compared to literature values. A strategy for the calibration of carboxyl-seminaphthofluorescein-1(c-SNAFL-1), a pH sensitive fluorophore, is discussed. Areas of application of the system are discussed and include probing intracellular pH, measurement of fluorescence resonance energy transfer (FRET), and measurement of pressure.
